Yes, you can certainly do that. It is a bit inelegant to dump a plate loading resistor, only to put it back in the form of a power supply dropping resistor (just my opinion of course). Interstage transformers also limit your choices of driver tubes to those with low plate impedance.I'm not really a fan of interstage transformers unless they are absolutely necessary, so I'm a bit biased.
