If it's a FIFO buffer, yes. Ideally, if the buffer truly eliminates transmission jitter, the differences in digital cables and transports should become inaudible.

But very few input buffers are FIFO- The better DACs often use PLLs, which do *not* seem to be totally immune to input jitter. (The DB Technologies DA924 DAC has a true FIFO buffer.)

And even if there is a FIFO buffer, differences in RFI bleedthrough amongst electrical cables will still bring audible differences. (The FIFO buffer still has a common ground, and the RFI just passes through the ground.) I think however, a FIFO buffer could make optical cables indistinguishable because RFI bleedthrough is non-existent. Provided the RFI generated from the optical converter isn't affected by input jitter.
