As Michael says, the amp should be optimized for the tubes in use. However, there can be another problem if you use 6550s in an amp designed for EL34s, and that is the grid-ground resistance. EL34s tolerate quite happily grid-ground resistors in the order of 0.5 Meg but with beam tetrodes like 6550, the grid-ground resistors need to be 100k or less to be stable.
