You can't simply measure it with a multimeter ...

on the Ohms range it will only measure the value of the input resistor, which will typically read several hundred K up to a meg. This resistor is in parallel with the tube's input capacitance, which not only varies depending on the type of circuit employed and the values used but reduces with frequency.

A typical 6SN7 grounded cathode input stage has an input impedance of 100 - 150K. This is measured at 20Khz, the accepted upper end of the audible spectrum. Compare this with the measured DC resistance measurement!

The input impedance can be easily predicted from a circuit diagram. Alternatively, you can measure it with a sig gen and a scope or an AC meter.
