Yes. Copying would be the simplest, albeit slowest way to backup the two drives. A file syncing utility such as Robocopy would be much better, as it could just copy whatever is either new or has changed, and delete whatever has been deleted. Or you could use backup software such as NTBackup that comes with Windows XP. For a music library, though, there's probably not much reason to do 'real' backups like that. If your library is encoded in a compressed format like Flac, one thing you should _not_ worry about is compressing the backup - it likely won't compress much more than it already has been.

I'm not familiar with iTunes, but maybe there's a way to backup the data and settings. You mostly need to worry about protecting the music files themselves, since they undoubtedly represent many hours of ripping CDs.
