How To Back Up 2x500G to 1TB Properly

I am using two 500 GB Lacie hard disks (named Lacie One and Lacie Two) for my music server using iTunes. What is the proper way to back up these two hard disks onto one TB Lacie hard disk. Is it as simple as creating two folders on the 1TB drive called Lacie One and Lacie Two and back up the two 500 GB disks into the respective folders? Obviously I want to be able to restore back into iTunes easily should one of the drives fail.
