IME, it is absolutely a no brainer to go with e-SATA over USB when one has a choice. I currently have a Toshiba laptop with e-sATA port and get at least 4x faster real-life transfer speeds via e-SATA over USB 2.0 to an external hard drive with both e-SATA and USB ports.

The technical max transfer speed of USB2 is never achieved with any of USB2 ports of my multiple laptops and desktops, and USB chips/external USB drives have a funny habit of developing master boot record errors and file system errors and eventually failing at some point. On the other hand, no hiccups with e-SATA, either with external e-SATA drive used for music storage or my another e-SATA drive which holds my OS as well as primary boot drive.
