It is indirectly in the signal path, as is any portion of the power supply.One can look at the output stage of a power amp as a mere gate or valve that regulates how much power supply goes to the output terminals, in that sense, the power supply IS the signal, just modulated by the output stage.
On the other hand, some amp designs have a large amount of PSRR or Power Supply Rejection Ratio, which measn that if the power supply has ripple riding on the DC voltage, that the ripple (hum) will not appear on the output very much. Some amps have a lot of PSRR, others have very little, and it depends on their exact topology.
How critical that cap is to the sound is a bit of a question, but my educated guess would be that it would not be nearly as much as all of those signal caps you replaced. It it likely there for HF bypassing of the main filter caps, to maintian low impedances at higher frequencies(often above the audio band)and those large caps will tend to dominate the sonic results anyway.
