I don't quite understand what tube you were testing, but if one of your tubes shorted that could cause the fuse to blow. If you take out the tubes and power it up does it blow the fuse? (don't touch the sockets) If the fuse still blows then I'd suspect the power supply rectifier and filter caps, but at this point if you don't know what you're doing take it to someone who does. I suppose it could be the standby switch itself - can you see any signs of arcing on it? If it doesn't blow the fuse with tubes out then get some new tubes and consider getting the amp biased.
