You would probably want to test them under the conditions they are running in the amp.
ARC should be able to provide that information.
I have an Amplitrex, and not all of the pre-programmed settings are correct.
Of course, you can make custom settings, which you may need to do for your particular operating conditions. Assuming you can computer control the Amplitrex (I have an old Windows Vista laptop for that and a Utracer), it doesn't look that hard to do (but I have never actually done it).
I can't answer your other questions, but on burn-in I think it is better if you can just run filaments for some period of time before applying other voltages.
