different for different tubes.

845s for example, the darkening comes from stray electrons that are hitting the inside of the glass. The darkening will be in a pattern, which can be seen to be the pattern made by the light (electrons) escaping from the filament where it is not blocked by plate/ceramic supports/etc.

866s and other MV tubes - same idea, they tend to get much darker with use, to the point of being black and unable to be seen through without strong light. something about the Mercury in them combined with the ionization and emission of electrons. Tbey dont' exhibit the shadowing effect, usually, that 845s and such do.

the darkening sometimes above the cathode on a small tube like 6sn7 is often getter migration, due to heat. usually small in area, and right above cathode/filament. also usually shiny, consistent with gettering materials moving up there.
