The end of almost any opera by Verdi, when performed well. Faves are Sutherland singing "Lassu nel ciel" from RIGOLETTO and Tebaldi singing the end of LA FORZA DEL DESTINO. Also, the moment in Act 2 of UN BALLO IN MASCHERA when Amelia first admits that she too is in love with Gustavus/Riccardo: "Ebben... si, t'amo." I could almost start crying right here in my cube as I write this.
Alfredo Kraus singing the end of Donizetti's LUCIA DI LAMMERMOOR. He's the only tenor I've experienced so far (on video, admittedly) who can make this scene the equal of, instead of a coda to, Lucia's mad scene. When he passed away recently, we watched the video from the Met in the early 1980s with Sutherland as a sort of tribute, and the tears welled up as soon as the curtain rose on the final scene. Knowing that Kraus's own wife had passed away only a short while before he did, hearing him sing Edgardo's lines about meeting his love again in heaven was heartbreaking.
