A speaker is designed to produce a signal with amplitude that varies with time. It is the amplitude that is a single value at every time. When you look at a varying amplitude over time, you can break it into frequency components. So the time varying signal consists of many frequencies in general. But frequency itself has no real meaning at a single instant, only over a time period. Amplitude at a single instant has a meaning and can be measured.
