I believe the 2A3 is the best all around tube for me. 45 is a little too low power & the 300B not quite the sonic quality of the 2A3 unless one goes with expensive new stock such as Emission Labs.
Designing an amp with high quality components around a tube is more important that the tube itself for best sonics. As for audio transformers, no inexpensive top quality parts exists.

Yes, the 45's sounds good as well but the low ouput power compared to 2A3's to me are a disadvantage. Of course, increase the speakers sensitivity and the 45's power is plenty but......noise becomes a very big factor!
To me, it is harder to solve system noise or hum issues in the amp than to build a new amplifier from trusted designs. Trying to figure out which components in your system contributes to the noise takes so much effort and experimentation.

My vote is for the 2A3 too. From a technical perspective, I think it's a great combination of output power, low plate impedance, and can be AC heated. Sonically I love the purity and "you are there" clarity it can provide.

My original audio DIY amplifier mentor, in 1982, described the 2A3 to me as the "direct access to the music" tube. Well said Robert, he positively hated 300Bs, BTW. RIP. He also always said, "chokes need to be 20 Ohms DCR or less". He'd love what I'm doing today !!

In 1982, I would also have hated 300Bs - there were only old-stock Western Electrics at outrageous prices, or a few crap copies, badly made and unreliable. The situation has improved immensely since then!
