Double check the voltages. B+ voltages for a power amp are normally in the 500 to 600 volt range for octal based power tubes. Since the unit came with the tube in, I would assume the previous owner was happy with the sound and you ought to be able to measure the rectified voltages without issue. If the voltages are high enough, there shouldn't be an issue as you can adjust the bias to cope. IIRC, without checking the manuals, the 5R4 is a higher impedance tube and should drop a few more volts than the 5Y3.
