Clarification: Fluffy insulation insufficient in standard wall?

I'm (finally) in the process of having a basement room drywalled. The new walls are going over block. The 2x4 studs are already up and are spaced at 16". The floor is concrete; ceilings rafters will remain exposed and packed with fluffy insulation (eventually).

My question: Can I use fluffy insulation behind the walls, or is there insufficient depth there to do any good? If not, I assume I should glue rigid fiberglass to the block...correct?

While I'm at it...given the stud spacing, I assume 1/2" drywall will be preferable to 5/8"?
