It was the solder joint. This is why we emphasize over and over that the first thing to do when there is a problem is reheat all the solder joints. 9 times out of 10 that will solve the problem.

The nature of DIY is often to 'undersimplify' when we have a troubleshooting issue that we haven't worked out. Always start with the basics like checking for correct wire connections, reheating to correct missing or cold solder joints and reseating tubes in their sockets before getting into more complex theories about what might be at issue.

It happens to everyone. You would not have wanted to be in the lab yesterday when I realized that I had miswired two wires on the Paramounts I am building - all 18 of them.
