Volume is loudness which is a combination of output and speaker efficiency. Gain in an amp is the ratio of change of input voltage to output voltage. In almost all amps(there are a very few exceptions) this is a constant ratio. Change in loudness is achieved by reducing the input voltage with a variable resistor, the volume control. The lower the setting of the volume control, the greater the resistance.
