A possibility is that it took some time for the cathodes to fully activate. Finally getting full emission will definitely alter behavior.

I've noticed that on tubes that haven't been used in decades it sometimes takes an apparently worn out tube a while to "wake up" and decide to emit electrons. Sometimes a few minutes, sometimes hours.
I can appreciate the burn-in process's effect on sonics, but which would be the best way to go about it?
- Run the tube as it should - with signal passing through?
- Run the tube simply with B+ applied?
- Any validity to the idea that you can burn-in quicker in a guitar amp where valves might be stressed a little harder?
Thanks in advance!

May your tubes be lively and long-lasting. Holy be thy heater.
Just plug the new tube in, make any necessary adjustments if required, and use the unit normally. Listen to some music, listen and enjoy as you get accustomed to the tone of the new tube and it does whatever its going to do in its early hours of use.
That's it.
