The Mark III differential output board can accommodate 6H30 tubes. However, to take advantage of the potential improvement, the operating point needs to be changed. The plate current should be changed to be 21mA and the cathode bias should be changed to be 5.5V.

Yes, to me it does make a very noticeable difference.
Some folks may call it a "tweak" but I call it a step-wise improvement. To me, the ability to make a series of small improvements over time keeps the circuit from becoming obsolete.
The difference between the Mark II active Output and the Mark IV Active Output is a series of small improvements, and the "Mark" designations are merely signposts along the road to sonic excellence.
One of the benefits of DIY is that you can try things, like using a different tube, and experience the effect of the change. Not all changes are positive so you must be open to learning what works and what doesn't work.
And even if you decide that you don't want to experiment, the older versions of the Active Output are still excellent performers.
