the filter that you plan to use after the rectifier and then a 10k ohm, 20 watt dummy load resistor you should be able to just turn the thing on....full voltage and see what happens.BTW 300vdc (a guess of what your target B+ is) / 10k ohms is 30ma. (again, a guess as to the current draw of your preamp)
These numbers should be close enough for you to see if the rectifier tube will work in your circuit and give you some kind of idea what it will do.
Running the power transformer on a variac adjusted low means the heater voltage to the rectifier tube will be low. Maybe low enough to keep it from working (conducting).
