Definitely depends on the power tube. IMO a power tube is "broken in" when its bias stops drifting in the amp. For example, I am currently breaking in a quad of Pissvane black bottle KT88 (the older series which have been replaced by the grey bottle versions). The company recommends 300 hours break-in time and I have had them in my amp for 3 weeks (maybe 100 hours playing time) and am still getting a fair bit of bias drift (5ma up or down). I check the bias every day or two when I use them.
Conversely, my first quad of EI KT90 seemed to have stable bias right from turn on and never drifted much. Same with the 6P3S-E tubes I got from Jim McShane, almost perfect bias from day one.
I definitely agree with the recommendation to only bias about 75% of normal on new output tubes until bias stops drifting. You might even like the sound at that bias level and want to stay there. Once bias is steady you can experiment.
