I don't remember offhand if the Missions are shielded. If not, they won't work well next to a CRT monitor. Other than that, they should sound ok on a desk, and for nearfield listening you probably won't need a subwoofer. But, as others have mentioned, it is unlikely a computer soundcard would be able to power them. Most likely you would need to go from the line out of the soundcard into an input of your amplifier to use those speakers. That's why pretty much all computer speakers have built-in amps.
As far as purpose-built computer speakers go, my favorites for sound quality are the Cambridge Soundworks 3 piece setups with the woofer box and cubes...
