I don't use a single reference disc because even well-made recordings vary in tonality and perspective, and because I try to listen to a variety of instruments with different demands, e.g., orchestra (the best test material because it challenges a system in the most ways), choral (a real torture test for nonlinear distortion), solo voice, pipe organ, and piano, which has unique demands and is also my personal "reference instrument" since I'm most familiar with it. I'll also listen to some multimiked recordings to get a feel for how the speakers handle recordings that are too bright, and, as I said, some pop recordings to see if the speakers will rock out.
That being said (and it's quite a mouthful), I do find that some recordings become "de facto" references. One that I use frequently is J. Gordon Holt's recording of the Delaware Symphony performing Jarnefelt's Praeludium on the first Stereophile test CD, made with two C-37's in ORTF configuration.
