The output from the cartridge is going to be cleaner if there are no speakers playing at the time the signal is digitized. Depending on your physical setup (room, position, vibration isolation, etc.) this may make a huge difference or it may be nearly immaterial. If you doubt this you can try recording the output of the cartridge with the TT powered down and while the speakers are playing some music. Or just footfalls on the floor...
